DAB preparation method

DAB (3,3'-diaminobenzidine, 3,3'-diaminobenzidine)
DAB is a substrate for the action of horseradish peroxidase. Its final product is a brownish-yellow precipitate, which is insoluble in organic solvents. It can be dehydrated by alcohol and xylene transparent to seal with neutral gum. Pay attention to the following points during use: 1. The DAB should be completely dissolved, and it can be used after filtering if necessary, otherwise the undissolved particles will be deposited on the specimen and affect the observation; 3. DAB has carcinogenic effects, so care should be taken during operation to avoid contact with the skin as much as possible.
1. DAB powder DAB (commonly used tetrahydrochloride) 50mg 0.05mol / L TBS (0.05M, pH7.6) 100ml 30% H2O2 30 ~ 40μl

Preparation method: first dissolve DAB with a small amount of TBS, then add the remaining amount of TBS, mix well to make the final concentration of DAB 0.05%, add 30% H2O2 30 ~ 40 after filtering, and filter out the precipitate That's it.
2. DAB kit (No. ZLI-9017)
Add 1 drop (about 50 μl) of Reagent 1 (DAB Concentrate) to 1 ml of Reagent 2 (DAB Buffer), mix well, and then prepare DAB working solution. This solution must be used as it is prepared. Store it in dark place after preparation. Use within 6 hours. The remaining liquid should be discarded. The biggest feature of this product is that it comes with a diluent, which avoids the impact of different water pH on DAB color development.
3. DAB kit (No. ZLI-9032)
Before use, just add one drop of each of the three reagents A, B, and C provided in the kit to 1 ~ 1.5ml of double-distilled water to obtain DAB working fluid, which is simple and easy to use.

Convenient folding for long battery life.
